A study published in BMCGastroEnterol finds that low-normal thyroid function is associated with an increased risk of advanced hepatic fibrosis in patients with metabolic dysfunction-associated fatty liver.

The univariable regression model did not detect any significant association between thyroid dysfunction and MAFLD risk. After adjusting for age and sex, the logistic regression analysis showed that low-normal thyroid function increased the risk of MAFLD . In the multivariable regression model additional adjusted for BMI, T2DM, and hypertension, the significant association between low-normal thyroid function and MAFLD disappeared.

The levels of TSH were significant difference between FIB-4 2.67 , NFS 0.676 . The univariable regression model showed that SCH and low-normal thyroid function were associated with FIB-4 values of > 1.30 and NFS values of > − 1.455 . These associations disappeared after adjusted covaries in the multivariable regression model. Low-normal thyroid function increased the risk of advanced fibrosis both in the univariable regression model and multivariable regression model.

MAFLD is a nomenclature proposed in 2020 to replace the definition of NAFLD. Therefore, most of the current studies have focused on elucidating the associations between thyroid function and NAFLD. Preimplantation genetic testing (PGT) is widely used to select euploid embryos for BRT carriers to increase the chance of a healthy live birth. Several strategies can be used to distinguish reciprocal translocation carrier embryos from those with a normal karyotype; however, these techniques are time-consuming and difficult to implement in clinical laboratories. In this study, nanopore sequencing was performed in two reciprocal translocation carriers, and the results were validated using the next-generation sequencing-based method named, “Mapping Allele with Resolved Carrier Status” (MaReCs). Results The translocation breakpoints in both reciprocal translocation carriers were accurately identified by nanopore sequencing and were in accordance with the results obtained using MaReCs. More than one euploid non-balanced translocation carrier embryo was identified in both patients. Amniocentesis results revealed normal karyotypes, consistent with the findings by MaReCs and nanopore sequencing. Conclusion Our results suggest that nanopore sequencing is a powerful strategy for accurately distinguishing non-translocation embryos from translocation carrier embryos and precisely localizing translocation breakpoints, which is essential for PGT and aids in reducing the propagation of reciprocal translocation in the population.

Association of shift work with incident dementia: a community-based cohort study - BMC MedicineAssociation of shift work with incident dementia: a community-based cohort study - BMC MedicineBackground Some observational studies had found that shift work would increase risks of metabolic disorders, cancers, and cardiovascular diseases, but there was no homogeneous evidence of such an association between shift work and incident dementia. This study aimed to investigate whether shift work would increase the risk of dementia in a general population. Methods One hundred seventy thousand seven hundred twenty-two employed participants without cognitive impairment or dementia at baseline recruited between 2006 and 2010 were selected from the UK Biobank cohort study. Follow-up occurred through June 2021. Shift work status at baseline was self-reported by participants and they were categorized as non-shift workers or shift workers. Among shift workers, participants were further categorized as night shift workers or shift but non-night shift workers. The primary outcome was all-cause dementia in a time-to-event analysis, and the secondary outcomes were subtypes of dementia, including Alzheimer’s disease, vascular dementia, and other types of dementia. Results In total, 716 dementia cases were observed among 170,722 participants over a median follow-up period of 12.4 years. Shift workers had an increased risk of all-cause dementia as compared with non-shift workers after multivariable adjustment (hazard ratio [HR], 1.30, 95% confidence interval [CI], 1.08–1.58); however, among shift workers, night shift work was not associated with the risk of dementia (HR, 1.04, 95% CI, 0.73–1.47). We found no significant interaction between shift work and genetic predisposition to dementia on the primary outcome (P for interaction = 0.77). Conclusions Shift work at baseline was associated with an increased risk of all-cause dementia. Among shift workers, there was no significant association between night shift work and the risk of dementia. The increased incidence of dementia in shift workers did not differ between participants in different genetic risk strata for dementia.
